چکیده

Skeletal muscle adapts to chronic physical activity by inducing mitochondrial biogenesis and switching proportions of muscle fibers from type II to type I. Several major factors involved in this process have been identified, such as the calcium calmodulindependent protein kinase IV (CaMKIV), calcineurin A (CnA), and the transcriptional component peroxisome proliferator-activated receptor coactivator 1 (PGC-1 ). Transgenic expression of PGC-1 recently has been shown to dramatically increase the content of type I muscle fibers in skeletal muscle, but the relationship between PGC-1 expression and the key components in calcium signaling is not clear. In this report, we show that the PGC-1 promoter is regulated by both CaMKIV and CnA activity. CaMKIV activates PGC-1 largely through the binding of cAMP response element-binding protein to the PGC-1 promoter. Moreover, we show that a positive feedback loop exists between PGC-1 and members of the myocyte enhancer factor 2 (MEF2) family of transcription factors. MEF2s bind to the PGC-1 promoter and activate it, predominantly when coactivated by PGC-1 . MEF2 activity is stimulated further by CnA signaling. These findings imply a unified pathway, integrating key regulators of calcium signaling with the transcriptional switch PGC-1 . Furthermore, these data suggest an autofeedback loop whereby the calciumsignaling pathway may result in a stable induction of PGC-1 , contributing to the relatively stable nature of muscle fiber-type determination.

Introduction: Nordic walking training has many benefits in improving the condition of the disabled elderly. Then the aim of this study was to evaluate the effect of Nordic walking training on plasma levels of PGC1α and SIRT6 in elderly women with diabetes. Materials and Methods: In this quasi-experimental study, 27 elderly women (age: 65.45±2.70 years) with type 2 diabetes were selected and ran...

Conjugated linoleic acid supplementation enhances insulin sensitivity and peroxisome proliferator-activated receptor gamma and glucose transporter type 4 protein expression in the skeletal muscles of rats during endurance exercise

Objective(s):This study examined whether conjugated linoleic acid (CLA) supplementation affects insulin sensitivity and peroxisome proliferator-activated receptor gamma (PPAR-γ) and glucose transporter type 4 (GLUT-4) protein expressions in the skeletal muscles of rats during endurance exercise. Materials and Methods:Sprague-Dawley male rats were randomly divided into HS (high-fat diet (HFD) s...

Compare the Effect of Eicosapentaenoic Acid and Oxidized Low-Density Lipoprotein on the Expression of CD36 and Peroxisome Proliferator-Activated Receptor Gamma

Background: There is evidence that CD36 promotes foam cell formation through internalizing oxidized LDL (ox-LDL) into macrophages therefore, it plays a key role in pathogenesis of atherosclerosis. In addition, CD36 expression seems to be mediated by nuclear receptor peroxisome proliferator-activated receptor gamma (PPAR-γ). The aim of the present study was to evaluate and compare the effect of ...

Role of peroxisome proliferator-activated receptor alpha and gamma in antiangiogenic effect of pomegranate peel extract

Objective(s): Herbal medicines are promising cancer preventive candidates. It has been shown that Punica granatum L. could inhibit angiogenesis and tumor invasion. In this study, we investigated whether the anti-angiogenic effect of pomegranate peel extract (PPE) is partly attributable to Peroxisome proliferator-activated receptors (PPARs) activation in the Human Umbilical Vein Endothelial Cell...

Butyrate supplementation to gestating sows and piglets induces muscle and adipose tissue oxidative genes and improves growth performance.

Weaned pigs often experience growth reduction immediately after weaning due to multiple stress factors associated with weaning. We tested the effect of prenatal and postnatal butyrate supplementation on growth performance of piglets. In study 1, piglets were orally gavaged with 0.3% butyrate from day 4 after birth to weaning (day 21). Butyrate increased ADG by 13% compared to saline treated con...
